At Wembley Stadium, Spurs will be 11-10 favourites, while BVB check in at 13-5. A draw comes in at 51-20 (all odds accurate as of Tuesday).
Both matches kick off at 8 p.m. GMT (3 p.m. ET) and will be available for broadcast via BT Sport (for UK viewers) and B/R Live (for U.S. viewers).
Madrid have found their best form of the season at a crucial time, winning six of their last seven matches across all competitions going into the first leg of their tie.
The only contest they failed to win was the Copa del Rey match against Barcelona, but that will also have been a boost for Los Blancos, as they bagged a draw at the Camp Nou and are in the driver's seat heading into the second leg.
The team finally appears to have found its footing under manager Santiago Solari, who has incorporated several youngsters and seems to have discovered the right balance.
Not everyone is excited about the direction of the La Liga giants, though:
While Los Blancos have discovered their best form this season, Ajax have gone in the opposite direction. They demolished a large portion of the Eredivisie clubs they faced in the first half of the campaign but have now lost two of their last three, including an upset away to Heracles.
The embarrassing 6-2 defeat against rivals Feyenoord highlighted some major defensive weaknesses that have surfaced following the departure of Maximilian Wober, who joined Sevilla in January. Madrid will relish the chance to take advantage.
Wednesday's other contest should be one of the tightest ties of the round. Hosts Spurs limped into the knockout phase of the tournament, and they will be aware they made it this far by the skin of their teeth:
Spurs have won three in a row after back-to-back defeats in domestic cup competitions and are alive again in the Premier League title race. While that's great news for the north London club, it adds extra strain to a squad that is already lacking in depth and can't afford to overlook any matches on the schedule.
The same holds true for Dortmund, who are winless in their last three matches and sit five points ahead of Bayern Munich in the Bundesliga.
Their squad depth is about to be severely tested, as BVB have been hit hard by the injury bug:
Both teams have an attacking philosophy, so fans could be in for a true spectacle at Wembley.
Predictions: Ajax 1-2 Real Madrid, Tottenham Hotspur 2-2 Borussia Dortmund
