The 34-year-old's solo blast traveled 485 feet at Coors Field.
Trout went 1-for-2 with one walk in the win.
He's now the second active MLB player with at least 400 career home runs, joining New York Yankees slugger Giancarlo Stanton.
Trout reached another important threshold earlier in the season, recording his 1,000th career RBI on July 27.
The three-time American League MVP's latest accomplishment is a silver lining on a 2025 season in which he hasn't been able to blow past expectations. Trout is hitting .229 with 22 homers, 13 doubles and 59 RBI while posting a .772 OPS.
He missed time earlier in the year due to a bone bruise in his left knee but has still appeared in 124 games, marking his highest total since the 2019 season.
Trout racked up 285 homers in the first nine years of his career, but injuries took a toll on his production. He appeared in just 36 contests during the 2021 season because of a right calf strain and failed to surpass 90 games played in two of the next three years due to more health issues.
Still, the 11-time All-Star has put together an incredible career in spite of his recent injuries and made history on Saturday.
