Tennessee pulled out to a 14-point lead in the first ten minutes of first-round action.
Wofford pushed back to come within eight points midway through the second half, but were unable to come any closer as the Volunteers cruised to victory.
Lanier, who finished the win with a game-high 29 points, shot 11-for-22 from the field and 6-for-13 from deep.
He is now just a pair of three-pointers shy of matching the all-time school record set by Chris Lofton in 2008.
Zakai Zeigler meanwhile contributing a 12-point, 12-rebound double-double while committing no turnovers in the win.
Zeigler also broke the all-time Tennessee career assists record on an assist to Lanier in the first half.
Tennessee will now advance to face the winner of UCLA and Utah State in the second round. Both of the team's All-SEC guards will be key to the Volunteers' hopes of making it past either opponent to qualify for the Sweet 16.
Lanier, Zeigler and the Volunteers will now wait to find out the identity of their next opponent after the first-round matchup between the Bruins and Aggies tips off Thursday night at 9:25 p.m. ET.
