McIlroy's victory put him over 1,100 FedEx Cup points ahead of Sepp Straka in second place.
The career Grand Slam holder won previously at the AT&T Pebble Beach Pro-Am and the Players Championship.
Sunday's win at Augusta National Golf Club was the second playoff victory for McIlroy this season. He also won in extra holes at The Players.
There's a long way to go between now and the end of the PGA Tour season, but McIlroy has established himself as the favorite to take home the title this summer.
Ludvig Aberg, Scottie Scheffler, Corey Conners and Justin Rose all made significant advances in the FedEx Cup standings with their performances at Augusta.
Rose took home 500 points to move up into ninth, Scheffler picked up 325 points to move six spots into fourth, Aberg's 250 points launched him up to third and Conners' 188 points kept him in sixth.
Sepp Straka and Russell Henley were the only golfers currently in the top 10 to not earn any FedEx Cup points at Augusta.
The next significant FedEx Cup points haul will come at the PGA Championship in May.
There are four weeks of PGA Tour play before the trip to Quail Hollow in Charlotte, starting with the RBC Heritage next week at Hilton Head Island, an event that Scheffler won last season.
