Gauff began her Roland-Garros run with a 6-4, 6-0 first-round victory over her compatriot.
Townsend gained an early 3-1 lead in the first set, but Gauff dominated down the stretch. Gauff went 11-1 in her last 12 games to seal the victory.
Gauff won the 2025 French Open, and her second Grand Slam singles title, with a three-set victory over Aryna Sabalenka.
The 22-year-old is now looking to become the first women's player since Poland's Iga Świątek, who won from 2022 to 2024, to repeat as champion in Paris.
