Koepka was unable to recover on Friday from a tumultuous first round at Royal Portrush, during which he hit two birdies but shot over par five times on the back nine.
This marks the third time in four major tournaments during which Koepka has missed the cut.
After finishing below the line at the Masters and PGA Championship, Koepka tied for 12th at the U.S. Open before struggling again at the British Open.
This also marks the third time in four years Morikawa has missed the British Open cut since winning the tournament in 2021.
Reed, who claimed a third-place finish at the Masters, and Griffin, who finished second this spring at the Memorial Tournament, were both unable to build on their recent success at Royal Portrush.
Scott struggled with three double bogeys in Friday's second round, while Cantlay made a late rally with a 12th-hole birdie before a bogey on Hole 15 spoiled his comeback.
It remains to be seen if Friday's results impact who U.S. Ryder Cup captain Keegan Bradley selects to join his the 2025 team. Competitors like Morikawa, Griffin, Cantlay and Koepka are currently ranked outside of the automatic Ryder Cup qualifying spots, but could still make the team as a captain's pick.
On the other side of the cut line, Scottie Scheffler's dominant 2025 season continued Friday as he topped the scoreboard by going 10-under through two rounds.
Matt Fitzpatrick, Brian Harman, Haotong Li and Rasmus Hojgaard rounded out the top five of the Open Championship standings heading into Saturday.
