Spain had also won the European Championships in 2008 and 2012.
As a result of the win for the South American side, Chile and the Netherlands advance into the Round of 16 before the two even face each other in Group B's final match.
Australia and Spain are stuck at the bottom of the group with zero points apiece.
Eduardo Vargas struck first for Chile, as he danced past Iker Casillas to finish off a brilliant move from Chile to give them a 1-0 lead.
Charles Aranguiz doubled Chile's lead and put Spain on the brink of elimination, hitting in a rebound off of a free kick just before half-time. A Chilean win will put the Netherlands and Chile through to the knockout stages before the two even face each other.
