With just two minutes left in the game, Harvard's Siyani Chambers was 1-of-9 from the field.
With 1:57 remaining, Chambers came alive with a clutch jumper to hand Harvard a three-point lead that it would not lose.
Chambers went on to make three of the final five Harvard free throws as well to hand Harvard its second upset in two years in the NCAA tournament.
Harvard was the better team throughout the entire game that featured plenty of shooting slumps by both teams.
Chambers (11), Laurent Rivard (11) and Wesley Saunders (12) were the only three Crimson players in double digits in the victory.
Cincinnati did receive a game-high 18 points from Sean Kilpatrick, but the star guard failed to come up big in the clutch for the Bearcats.
Harvard will face either No. 4 Michigan State or No. 13 Delaware on Saturday in the third round in Spokane.
